The U's fought back from 1-0 down to take all three points at Kenilworth Road.
The home side took the lead through Andrew Fotiadis but conceded an equaliser just before half-time - Karl Duguid firing home a free-kick.
Luton had dominated the first half with Simon Brown saving well from Fotiadis and Robinson.
But after the break Colchester came more into the match and striker Dean Morgan scored the winner - a header from Kem Izzet's cross which gave goalkeeper Mark Ovendale no chance.
