A first-half penalty from Macclesfield Town striker Lee Glover was enough to beat Southend United at the Moss Rose Ground on Saturday.
The spot-kick was awarded in the 29th minute when Blues left-back Damon Searle was adjudged to have fouled Richard Tracey in the Shrimpers' box.
Southend came close to equalising on a number of occasions, with home goalkeeper Steve Wilson pulling off a tremendous save to deny Southend substitute Mark Rawle late in the game.
